What is  Cyclophosphamide  ?

Cyclophosphamide is one of the most commonly used alkylating antitumor agents. It is white crystalline or crystalline powder at room temperature (the loss of crystalline water is liquefied) and stable at room temperature. Soluble in water, but not very soluble. Aqueous solution is unstable, so it should be used shortly after dissolution. Soluble in ethanol. First synthesized in 1958 by Arnold and Bourseaux, the synthesis of this compound and its derivatives is based on the structure and action of previous anticancer drugs. Mainly used for tumor immunity, after entering the human body, in the liver microsomal enzyme catalyzed to explain the alkylation of strong chloroethylphosphamide (or phosphamide nitrogen mustard), and produce cytotoxic effect on tumor cells, has obvious inhibitory effect on a variety of tumors, in recent years because it has been proved to have immunosuppressive effect and used in the treatment of a variety of autoimmune diseases, has achieved obvious efficacy.

Fuction and Application of   Cyclophosphamide 

Cyclophosphamide (CTX) is a cell cycle non-specific cytotoxic drug, but it has a stronger effect on G2 phase (late DNA synthesis).
The main immunosuppressive mechanism is as follows:
The absolute number of T and B lymphocytes decreased, and the early B lymphocytes were more obvious.
Inhibition of lymphocyte to specific antigen stimulation of the mother cell transformation. The inhibition of mitogen stimulation was
not as good as that of specific antigen.
Inhibition of antibody response to neoantigens and delayed skin allergy.
Decrease the elevated immunoglobulin level, long-term use (several years) after the possible development of hypopromaglobulinemia.
In vitro selective inhibition of B-lymphocyte function, reduce the spontaneous production of immunoglobulin by some B-lymphocytes,
and inhibit the production of immunoglobulin by general mitogen stimulation. Cyclophosphamide has no cytotoxic effect before metabolism,
and its activation depends on the oxidation of cytochrome P450 in the microsomal enzyme system of liver cells to form
4-hydroxycyclophosphamide, which becomes highly active phosphamide nitrogen mustard after ring-opening, thus exerting cytotoxic effect.
