11-alpha-Hydroxycarvenone is obtained from canrenone by microbial transformation of the hydroxyl group at the 11 position. In 1971, B. lunt et al. tried to produce 11α?hydroxycannarone from canrenone. in 1980, Deshayes achieved 95% conversion at a canrenone feed of 1.5 g/L, but the large feed volume made it unsuitable for large-scale production. in 1996, Ng; John S. et al. Intermittent replenishment was used, and although the feeding volume was greatly increased, the conversion time was lengthened and the conversion rate was not high, resulting in a much higher cost.11α-Hydroxycannarone is a steroidal derivative formed after the hydroxylation reaction of canrenone, and it is also an important pharmaceutical intermediate for the synthesis of eplerenone. As a novel drug for the treatment of hypertension and other cardiovascular diseases, eplerenone has a broad Chemicalbook market prospect. The method of microbial conversion of canrenone for the synthesis of 11α-hydroxycannarone has the advantages of high specificity, mild reaction conditions, high catalytic efficiency, few by-products, low cost and low pollution. Due to the extremely low solubility of 11α-hydroxycannarone in water, 11α-hydroxycannarone synthesized by microbial fermentation is mainly adsorbed on the surface of mycelium, which is difficult to be separated by purely physical methods such as filtration, centrifugation or extraction. At the same time, there are also unconverted canrenone in the fermentation broth, and the physical and chemical properties of canrenone are similar to those of 11α-hydroxy canrenone, so the purification of the extract is also an important step. The existing microbial conversion technology of canrenone still suffers from long production cycle, high cost and low conversion rate, which greatly limit the application and promotion of this product.
